Mercer received national attention in 2014 when the men's basketball team defeated Duke University in the second round of the NCAA Tournament; the team was defeated by the University of Tennessee in the third round. The team finished the season with a 27–9 record, Mercer's third straight with more than 20 victories. In honor of its win over Duke, Mercer received the 2014 Espy Award for Best Upset.

On November 19, 2010, Mercer announced the reinstatement of intercollegiate football beginning in the fall of 2013. The university competed as an NCAA Division I, non-scholarship program in the Pioneer Football League in 2013, and is now a scholarship program in the Southern Conference.
